General description

GRB2-related adapter protein 2 (UniProt O75791; also known as Adapter protein GRID, GRAP-2, GRB-2-like protein, Grb2-related adapter protein downstream of Shc, GRB2L, GRBLG, GRBX, Grf-40, Grf40 adapter protein, Growth factor receptor-binding protein, Hematopoietic cell-associated adapter protein GrpL, P38, Protein GADS, SH3-SH2-SH3 adapter Mona) is encoded by the GRAP2 (also known as GADS, GRB2L, GRID) gene (Gene ID 9402) in human. GADS (Grb2-related adapter protein downstream of Shc), GRB2 and GRAP constitute the GRB2 family of adapter proteins characterized by a central SH2 domain (a.a. 58-149 of GADS) flanked between two SH3 domains (a.a. 1-56, 271-330 of GADS). Although the SH2 domains of all three members have similar binding specificities, the C-terminal SH3 domain of GADS possesses unique binding specificity that allows for its interaction with the adapter protein SLP-76 (SH2 domain-containing leukocyte protein of 76R01;kDa). The GADS-SLP-76 complex is recruited to tyrosine-phosphorylated LAT (linker for activation of T cells) via GADS SH2 domain, allowing the assembly of signaling complexes critical to T-cell activation. In addition, evidences indicate that GADS plays a critical role in the development of BCR-ABL-mediated lymphoid disease. The oncogenic Bcr-Abl fusion protein is shown to bind GADS in a phosphotyrosine-dependent manner and GADS is expressed in a subset of CML cell lines and B-ALL patient samples, where it associates with both BCR-ABL and SLP-76.

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected GADS Thr262 phosphorylation induction in 10 µg lysate from Calyculin A- and TNFα-treated Jurkat cells.

Specificity

Clone 6E11.2 detects GADS phosphorylated on Thr262. The corresponding phosphorylation site in human GADS spliced isoform 2 is Thr149 (UniProt O75791-2).

~40 kDa observed. 37.91 kDa (isoform 1) and 24.38 kDa (isoform 2) calculated. Uncharacterized band(s) may appear in some lysates.
